Anti-inflammatory effects of tirzepatide: a systematic review and meta-analysis

Abstract
Tirzepatide, a dual glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptor agonist, has demonstrated substantial metabolic and weight-reduction benefits; however, its anti-inflammatory effects remain less established. This systematic review and meta-analysis, conducted according to PRISMA guidelines, included randomized clinical trials and observational studies evaluating changes in high-sensitivity C-reactive protein (hsCRP) and interleukin-6 (IL-6). A random-effects model was applied. Tirzepatide significantly reduced hsCRP and IL-6 levels compared with placebo across different doses and study populations. These findings suggest that tirzepatide exerts significant anti-inflammatory effects, potentially contributing to its broader cardiometabolic benefits.
